Abstract

Metabolic stress and lipid peroxidation are key complications in the damage associated with type 2 diabetes. The aim of this study was to investigate the effect of moderate-intensity aerobic training and herbal compound Plantamajoside on plasma FGF21 and MDA responses in type 2 diabetic rats. In this experimental study, 30 male C57bl/6 mice (3-4 months, weight 24 ± 2 g) were divided into five groups: healthy control, diabetic control, diabetic-Plantamajoside, diabetic-aerobic exercise, and diabetic-aerobic exercise-Plantamajoside. The supplementation groups received 100 mg of Plantamajoside (per kg body weight) diluted in distilled water orally daily during the intervention period. The incremental moderate-intensity exercise protocol was performed on treadmill at 70% of maximum oxygen consumption, for 45 minutes per day, 5 days per week, for 8 weeks. After anesthesia, autopsy was performed, and blood samples were collected. The Metabolic stress indices (FGF21) and lipid peroxidation (MDA) were measured. Data were analyzed using two-way analysis of variance, and Tukey's post hoc test at the P<0.05. The results showed that moderate-intensity aerobic training had significant effect on plasma FGF-21 concentration (F=5.583, P=0.026) and plasma MDA concentration (F=9.394, P=0.005). The herbal supplement Plantamajoside had no significant effect on plasma FGF-21 concentration (F=0.125, P=0.726) and plasma MDA concentration (F=0.546, P=0.466). Also, the interaction between moderate-intensity aerobic training and Plantamajoside had no significant effect on plasma FGF-21 concentration (F=2.632, P=0.117) and plasma MDA concentration (F=2.017, P=0.167). According to the results, moderate-intensity aerobic training can help reduce metabolic stress and lipid peroxidation in type 2 diabetic rats.
