چکیده

زمینه و هدف: ورزش از طریق تنظیم برخی از RNAهای غیرکدکننده، اثرات محافظتی قابل توجهی در برابر اختلال عملکرد اندوتلیال دارد. بنابراین، هدف از این مطالعه بررسی مکانیسم زمینه‌ساز بهبود اختلال عملکرد اندوتلیال توسط ورزش در نوجوانان دارای اضافه وزن و چاق بود.
مواد و روش‌ها: 40 نوجوان پسر با دامنه سنی 12-15 ساله، دارای اضافه وزن و چاق انتخاب شدند و به‌صورت تصادفی در دو گروه تجربی و کنترل (20 نفر در هر گروه) تقسیم شدند. گروه تجربی به‌مدت 12 هفته تمرینات ترکیبی (تناوبی با شدت بالا و مقاومتی) را سه جلسه در هفته اجرا کردند و گروه کنترل در هیچ برنامه ورزشی شرکت نکرد. شاخص‌های آنتروپومتریک، سطوح پروفایل لیپیدی، اتساع وابسته به جریان (FMD)، نیتریک اکسید (NOx) و میکرو آر ان آ-126 (miR-126) پیش و پس از مداخله، اندازه‌گیری شد.
نتایج: پس از 12 هفته مداخله، سطوح FMD، NOx و miR-126 در گروه تمرین، افزایش معنی‌داری نشان داد (05/0≥p). همچنین، تمرینات ترکیبی موجب بهبود معنی‌داری در شاخص توده بدنی (BMI)، وزن بدن و درصد چربی بدن در گروه تمرین شد (05/0≥p). به‌علاوه، نتایج نشان داد بین تغییرات miR-126 وBMI همبستگی منفی و معنی‌داری وجود دارد (001/0=P، 951/0-=r)؛ درحالی‌که، بین تغییرات miR-126 و FMD (001/0=P، 771/0=r) و بین تغییراتFMD و NOx همبستگی مثبت و معنی‌داری گزارش شد (001/0=P، 896/0=r).
نتیجه‌گیری: نتایج نشان می‌دهد تمرینات ترکیبی می‌تواند با تغییرات miR-126 و کاهش وزن در نوجوانان دارای اضافه وزن و چاق، اثرات محافظتی قابل توجهی در اختلال عملکرد اندوتلیال داشته باشد

The effect of combined high-intensity interval training (HIIT) and resistance training on miR-126 expression and its relationship with vascular endothelial function in overweight and obese adolescents

Background and Objective: Exercise has significant protective effects against endothelial dysfunction through the regulation of some non-coding RNAs. Therefore, the aim of this study was to investigate the underlying mechanism of improvement of endothelial dysfunction by exercise in overweight and obese adolescents. Materials and Methods: 40 overweight and obese adolescent boys aged 12-15 years were selected and randomly divided into two experimental and control groups (20 subjects in each group). The experimental group performed combined exercises (high-intensity interval training and resistance) three sessions per week for 12 weeks, and the control group did not participate in any exercise program. Anthropometric indices, lipid profile levels, flow-dependent dilation (FMD), nitric oxide (NOx), and microRNA-126 (miR-126) were measured before and after the intervention. Results: After 12 weeks of intervention, FMD, NOx and miR-126 levels in the exercise group showed a significant increase (p≥0.05). Also, combined exercises significantly improved body mass index (BMI), body weight and body fat percentage in the exercise group (p≥0.05). In addition, the results showed that there was a negative and significant correlation between miR-126 changes and BMI (P=0.001, r=-0.951); while, there was a positive and significant correlation between miR-126 changes and FMD (P=0.001, r=0.771) and between FMD changes and NOx (P=0.001, r=0.896). Conclusion: The results show that combined exercises can have significant protective effects on endothelial dysfunction by changing miR-126 and weight loss in overweight and obese adolescents.
