چکیده

هدف: دیابت منجر به فیبروز قلبی شده که با پیشرفت بیماری بدتر می‌شود و به کاردیومیوپاتی دیابتی منجر می‌شود. در ا ین مطالعه اثر فعالیت ورزشی هوازی (AT) همراه با بربرین کلراید (BR) بر بیان TGF-β وSmad4 بافت قلب موش‌های صحرایی دیابتی بررسی شد.
روش شناسی: تعداد 32 موش صحرایی نر ویستار (03/17±09/276 گرم، 10 هفته‌ای) به طور تصادفی به چهار گروه (8=n): دیابت (DM)، دیابت -بربرین (BRDM)، دیابت-تمرین هوازی (ATDM)، دیابت-تمرین هوازی-بربرین (ATBRDM)، تقسیم شدند. دیابت با تزریق استرپتوزوتوسین القا شد. گروه‌های تمرین به مدت شش هفته برنامه تمرین هوازی فزاینده (18-10 متر در دقیقه، 40-10 دقیقه در روز، پنج روز در هفته) را روی تردمیل انجام دادند. بربرین کلراید (mg/kg/day 30) یک بار در روز به صورت خوراکی با گاواژ خورانده شد. در پایان هفته ششم نمونه بافت قلب جمع‌آوری شد. سطوح بیان ژنی TGF-β و TGF-β به روش real- time PCR اندازه‌گیری شد. داده ها با استفاده از آزمون ANOVA در سطح معنی‌داری 05/0 p< آزمون شد.
یافته‌ها: نتایج نشان داد که AT، BR و ترکیب AT-BR در موش‌های دیابتی باعث کاهش معنی‌دار در میزان TGF-β1 (0001/0=p) وSmad4 (0001/0=p) بافت قلب شد. همچنین کاهش معنی‌داری در میزان این شاخص‌ها در گروه ATBRDM نسبت به BRDM و ATDM مشاهده شد (05/0 p<).
نتیجه‌گیری: AT و BR با بهبود نشانگرهای فیبروز بافت قلب، یک استراتژی موثر برای بهبود فیبروز قلبی در موش‌های دیابتی با STZ می‌باشد. علاوه بر این، AT و BR اثر هم‌افزایی محافظتی قلبی را برای کاهش نشانگرهای فیبروز قلبی نشان داد.

Interaction effect of Aerobic Training and Berberine Chloride intake on the Expression of TGF-β and Smad4 of Heart Tissue in Streptozotocin-Induced Diabetic Rats

Aim: Diabetes leads to extensive cardiac fibrosis, which worsens as disease progresses, culminating in diabetic cardiomyopathy. In this study, we investigated the effect of aerobic exercise (AT) and berberine chloride (BR) on the expression of TGF-β/Smad4 heart tissue in diabetic rats.
Methods: In this experimental study, 32 male Wistar rats (276/09±17.03, 10 weeks old) were randomly divided into four groups (n=8): Diabetes (DM), Diabetes-Berberine (BRDM), Diabetes-Aerobic Training (ATDM), Diabetes-Aerobic Training-Berberine (ATBRDM). Diabetes was induced by injection of streptozotocin. Training groups have performed a progressive aerobic running program (at 10-18 m/min, 10-40 min/day, and 5 days/week) on a motor-driven treadmill for six weeks. Berberine chloride (30 mg/kg/day) were administered orally, by gavage, once a day. At the end of six weeks-- heart tissue samples were collected. TGF-β and Smad4 gene expression levels were measured by real-time PCR. Data were analyzed using ANOVA at p<0.05.
Results: The results showed that AT, BR and AT-BR combination in diabetic rats significantly decreased TGF-β (p = 0. 0001)) and Smad4 (p = 0. 0001) in heart tissue. Also, a significant decreased the amount of this index was observed in the ATBRDM compared to the BRDM and ATDM groups (p <0.05).
Conclusions: AT and BR by improving heart tissue fibrosis markers is an effective strategy to improve cardiac fibrosis in diabetic rats with STZ. In addition, AT and BR supplementation showed a protective cardioprotective effect of reducing cardiac fibrosis markers.
