چکیده

مقدمه: هدف از تحقیق حاضر بررسی اثر مصرف عصاره گزنه و تمرینات هوازی بر بیان ژن VEGF و کاسپاز 3 در موش های مبتلا به سرطان ملانوما بود.
روش شناسی: در این تحقیق 20 سر موش صحرایی نر بالغ هشت هفته ای به صورت تصادفی به 4 گروه شامل گروه‌های: کنترل ، تمرین، عصاره و تمرین+عصاره تقسیم شدند. برنامه تمرین شامل 30 دقیقه دویدن روی تردمیل بدون شیب و با سرعت 16 متر در دقیقه برای هفته اول بود و هر هفته یک متر بر دقیقه اضافه شد تا در هفته هشتم به 22 متر بر دقیقه رسید. یک هفته پس از القا سرطان ملانوما، گروه تجربی میزان 30 mg/kg/day عصاره اتانولی گیاه گزنه را به روش خوراکی و به مدت 8 هفته مصرف کردند. برای اندازه گیری میزان بیان ژن VEGF و کاسپاز 3 از روش RT PCR در خون استفاده شد.

یافته ها: تجزیه و تحلیل داده ها نشان داد بیان ژن VEGF در گروه های تجربی نسبت به گروه کنترل روند کاهشی داشت؛ ولی به سطح معناداری نرسید(0.154p= ). همچنین نتایج نشان داد که بیان ژن کاسپاز3 در گروه های تجربی نسبت به گروه کنترل افزایش معناداری داشت(0.000=p).همچنین آزمون تعقیبی نشان داد که بیان ژن کاسپاز3 بین گروه ترکیبی با سایر گروهها نیز تفاوت معناداری وجود دارد.
بحث: نتایج نشان داد که احتمال دارد مصرف عصاره گزنه از طریق مکانیسم های مختلف مانند فعالیت آنتی اکسیدانی، القاء آپوپتوز جلوگیری از رشد سلول و مهاجرت سلولها اثرات ضد سرطانی ایجاد کنند.

Introduction: The aim of this study was to evaluate the effect of nettle extract consumption and aerobic exercise on VEGF and caspase 3 gene expression in mice with melanoma.
Methodology: In this study, 20 adult male rats with age of 8 week were randomly divided into 4 groups including: control, exercise, extract and exercise + extract. The training program consisted of 30 minutes of running on a treadmill without a slope at a speed of 16 m/min for the first week, and one meter per minute was added every week until it reached 22 meters per minute in the eighth week. One week after melanoma induction, the experimental group consumed 30 mg /kg /day of nettle ethanol extract orally for 8 weeks. RT PCR in blood was used to measure the expression of VEGF and caspase 3 genes.

Results: Data analysis showed that VEGF gene expression was decreased in experimental groups compared to the control group; But did not reach a significant level (p = 0.154). The results also showed that caspase 3 gene expression was significantly increased in the experimental groups compared to the control group (p = 0.000). Post hoc test also showed that caspase 3 gene expression was significantly different between the combination group and other groups.
Conclusion: The results showed that consumption of nettle extract may have anti-cancer effects through various mechanisms such as antioxidant activity, induction of apoptosis inhibition of cell growth and cell migration.
